Abstract: This study investigated the transgenerational neurotoxic effects of DEHP on zebrafish and the epigenetic mechanism by model construction, behavioral testing, and molecular biology. we used the molecular descriptor information of the compounds to construct a prediction model of the criteria parameters by multiple linear regression, and derived the water quality criteria value of DEHP as 40.85 ug/l. Using this value as a reference to determine the concentration gradient, we exposed the zebrafish to DEHP solution for 30 days to establish the parental model. Localized necrosis of brain tissues in the exposed group was observed by histopathological methods; epigenomic results showed that the genomic methylation level of brain tissues in the exposed group was significantly reduced. The findings revealed that DEHP exerts epigenetic neurotoxicity, reducing the DNA methylation level in zebrafish brain tissues and consequently leading to the inheritance of neurological damage and protein secretion disorders. The study also showed that low concentrations of DEHP still have heritable neurotoxic effects on zebrafish, which provides theoretical references for benchmarking studies of DEHP and environmental risk management studies.
